Uma equipe de pesquisa liderada pela Universidade da Califa³rnia, Irvine, descobriu novos circuitos neurais que regulam o aprendizado espacial e a memória na formação hipocampal do cérebro. A equipe identificou novos papanãis funcionais de novas conexões de circuito entre a regia£o CA1 venal e as regiaµes CA3 dorsais do hipocampo e demonstrou que a inativação genanãtica dessa projeção prejudica o aprendizado espacial e a memória relacionados ao objeto, mas não modula os comportamentos relacionados a ansiedade.
O estudo, intitulado "Projeções não cana´nicas para o hipocampo CA3 regulam o aprendizado espacial e a memória, modulando a via trissina¡ptica do hipocampo feedforward", foi publicado hoje na PLOS Biology .
O hipocampo não éuma área cerebral homogaªnea. O eixo septotemporal, ao longo do qual estãolocalizada a via trissina¡ptica, separa a regia£o dorsal do hipocampo, que estãomais envolvida na aprendizagem, memória e navegação espacial, e a regia£o ventral, que desempenha um papel no comportamento emocional. A organização do circuito unidirecional e feedforward do trissina¡ptico estãobem documentada, mas a conectividade entre as regiaµes septais e temporais émenos bem descrita.
"Nossas descobertas estendem o conhecimento da conectividade hipocampal e sua relação com os processos de aprendizagem e memória em todo o eixo septotemporal e fornecem uma base de circuito para explorar esses novos papanãis funcionais", disse Xiangmin Xu, Ph.D., Chancellor's Fellow e professor de anatomia e neurobiologia e diretor do Centro de Mapeamento de Circuitos Neurais (CNCM) da Faculdade de Medicina da UCI. “O novo mecanismo do circuito hipocampal éaltamente relevante para o tratamento de distúrbios de aprendizagem e memória, incluindo a doença de Alzheimerâ€.
Com base em seu trabalho anterior, Xu e sua equipe usaram vários marcadores virais, incluindo rastreamento retra³grado de raiva monossina¡ptica e rastreamento antera³grado baseado em herpes (H129) para estabelecer novas projeções de CA1 do hipocampo para CA3. Resultados de mapeamento robustos mostraram que as entradas de CA1 para CA3 correm opostas a via trissina¡ptica e em uma direção temporal para septal. Eles também descobriram que a inativação genanãtica da projeção CA1 a CA3 prejudicou a aprendizagem espacial e a memória relacionadas ao objeto, mas não modulou os comportamentos relacionados a ansiedade.
"O surgimento de técnicas de mapeamento genanãtico viral aumenta nossa capacidade de determinar a complexidade detalhada do circuito cerebral", disse Xu. "Nosso estudo foi possível graças a s novas ferramentas genanãticas virais desenvolvidas por nossos pesquisadores do CNCM na UCI.
